John was born free in 1829 in Virginia, the mixed race child of a freedwoman of mixed race (African American and Native American heritage) named Lucy Jane Langston and a white planter from England named Ralph Quarles. Ralph arranged for his Quaker friend, William Gooch, to become the guardian of his and Lucy's children.
